摘要
目的:观察骨髓基质细胞的培养及不同浓度碱性成纤维生长因子(bFGF)对其生长的影响。方法:采用全麻下抽取兔股骨骨髓,经体外培养的方法并在培养液中加入4种不同浓度的bFGF(0,60,120,240ng/ml)培养后经倒置显微镜观察;并用试剂盒测定细胞碱性磷酸酶的含量。结果:60、120ng/ml的bFGF能明显促进骨髓基质细胞的生长(P<0.01),其余两组无明显差异。而4组之间的细胞碱性磷酸酶含量在3天时无明显差异,6天时加入bFGF组的细胞碱性磷酸酶含量明显下降。结论:bFGF对骨髓基质细胞的影响是复杂和多方面的;不同剂量浓度对骨髓基质细胞生长的影响亦不一样,并不呈正相关,合适的有效浓度为60~120ng/ml。
Obejective:To investigate the method of culture bone marrow stromal(BMSC) and the effects of basic fibroblast growth factor (bFGF) on proliferation of BMSC.Methods: The BMSC was cultured in vitro with different concentrations of bFGF (0,60,120,240 ng/ml).The speed of cell growth were observed under inverted microscope and the cell alkaline phosphatase(AKP) was measured with the AKP kit.Results:The concentration of 60,120 ng/ml bFGF could significantly promote cell proliferation and cellular AKP activity in four groups had no significant differences. Conclusion:The bFGF may have compliex biological effects on BMSC.The effects are dependent on concentration.Its available concentration is 60-120 ng/ml.
