卵巢癌患者红细胞CR_1密度基因型与红细胞免疫粘附活性的相关研究 被引量:1

摘要 目的:探讨卵巢癌患者红细胞补体受体Ⅰ型(E-CR1)密度基因型与红细胞免疫粘附活性的关系。方法:采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法检测E-CR1密度基因型,采用红细胞免疫粘附试验测定红细胞CR1(E-CR1)粘附肿瘤细胞活性、粘附免疫复合物数量(E-ICR)。结果:卵巢癌患者E-CR1密度基因多态性分布为纯合子HH型50.0%,杂合子HL型44.1%,纯合子LL型5.9%,与正常对照组(纯合子HH型83.3%,杂合子HL型16.7%,纯合子LL型0%)比较有显著差异(P<0.01);HH型及HL型患者的E-CR1粘附肿瘤细胞活性[(30.43±4.67)%和(24.15±3.82)%]均低于正常对照组[(38.76±5.34)%和(30.51±5.46)%](P<0.01和P<0.05),E-ICR则高于正常对照组(P<0.01)。结论:卵巢癌患者E-CR1密度基因型与红细胞免疫粘附活性密切相关。 ve: To study on the relationship between density genotypes of erythrocyte complement receptor type 1 (E-CR1) and erythrocyte immune adherence activity in the patients with ovarian cancer. Methods: Genomic determination of E-CR1 was performed using polymerase chain re-action-restriction fragment length polymorphism ( PCR-RFLP) . Erythrocyte immune adherence test was used to determine the activity of E-CR1 adhering to tumour cells and the rate of erythrocytes ad-hering to immune complex (E-ICR) . Results: The percentages of genomic density polymorphism distributions in patients with ovarian cancer(homozygous HH:50.0%, heterozygous HL:44.1%,ho-mozygous LL:5.9%) were significantly higher than those in control group (HH:83.3% ,HL: 16.7% , LL:0%)(P<0.01). The adhering activities of E -CR1 to tumor cells in patients were (30.43±4.67)% and (24.15±3.82)% for HH and HL types respectively,while the adhering activities of E-CR1 to tumour cells in control group were (38.76±5.34)% and (30.51±5.46)% respectively (P < 0.01 and P < 0.05). Conclusion:There may be obviously correlation between E-CR1 density genotypes and erythrocyte immune adherence activity in patients with ovarian cancer.
