摘要
本研究在已制备的 AEV单抗基础上建立了检测 AEV抗原的直接 Dot- ELISA方法。该方法对 AEV最低检出量为 8.0 3μg.m L-1。人工感染 1日龄 SPF鸡 ,取临床发病鸡 (6~ 1 3日龄 )相应病料 ,各病料样本病原阳性检出率分别为 :脑 98% ,胰 94% ,十二指肠 80 % ,腺胃 85%。对 1 0 0份临床自然发病鸡病料检测 ,阳性检出率为 90 %。对 1 0份 AEV病原分离阳性样品检测阳性率为 1 0 0 %。
The procedure of direct Dot ELISA has been successfully established for detection of AEV antigen, and the detection limits of the AEV was 8.03μg·mL -1 , The one day old SPF chickens were infected with AEV (VR and Shandong isolates) by the way of brains and subcutaneous in jection in neck, and the pathological tissues and organs infected were collected when they are 6~13 day old. It was shown that the positive rates of AEV antigen in brain, pancreas, duodenum, gland stomach are 98%, 94%, 80%, 85% respectively. These organs are the major targets infected by AEV. One hundred samples collected from field infected chickens having clinical sign were detected, and the positive rate of AEV was 90%.
