摘要
目的观察顺铂内耳中毒豚鼠耳蜗热休克蛋白70(HSP70)的表达特点。方法制备顺铂内耳中毒听力损伤模型,分别于第3天,第6天处死豚鼠,取耳蜗做石蜡包埋切片,进行HSP70免疫组化(SP法)染色,检测耳蜗HSP70的表达和分布特点。图像分析采用计算机图像分析系统。结果对照组中Corti器、血管纹、螺旋缘、螺旋神经节呈弱阳性。Ⅱ组中Corti器、血管纹、螺旋缘、螺旋神经节的HSP70表达增强。Ⅲ组中Corti器、血管纹、螺旋缘、螺旋神经节呈强阳性。Ⅲ组平均灰度值较对照组有显著性差异(P<0.01)。结论顺铂能够诱导HSP70在豚鼠耳蜗中的表达,使HSP70在豚鼠耳蜗中的表达增强。
Objective To study the expression of the heat shock protein 70 in Guinea Pigs cochlea after cisplatin(DDP) ototoxicity.Methods Preparing the animal model of DDP-induced hearing loss.After successive 3 and 6 days respectively,all animals were sacrificed.Cochlea were taken for preparation cochlea section.The expression of HSP70 was detected by SP immunohistochemistry method and averagegray densities were quantified with a light microscope image analysis system.Results HSP70 was presentin the control group at a lower level,including Corit sorgan,stria vascularis,spiral limbus,spira lganglion cells.The level of HSP70 immunoreactivity in the guinea pig cochlea of DDP group animal were high,including Cortis organ,stria vascularis,spiral limbus,spiral ganglion cells.especially after successive 6 days.The average gray densities of HSP70 of DDP group animal were significantly different(P<0.01)from the control group.Conclusion DDP can induce the expression of HSP70 in Guinea Pigs cochlea.
