摘要
目的 :分析受乙肝疫苗保护后部分儿童的“免疫无效”现象。方法 :采用巢式 PCR- SSCP银染技术对 6 0例曾受疫苗保护的儿童血清中 HBV DNA S基因片段进行研究。结果 :在 HBs Ag阴性血清中存在 HBV S基因点突变 ,该突变位于从 HBV基因组 Eco R 位点算起的第 36 7~ 36 9核苷酸区域内。结论 :肝癌高危儿童中发现存在 HBV逃脱变异。疫苗刺激机体所产生的抗体不能中和入侵的 HBVa决定簇 ,也不能阻断 HBV感染致使产生“免疫无效现象”。
Objective:To analyse the 'immune null and void phenomenon' among part children protected by injecting HBV vaccine.Methods:Using nest PCR-SSCP Ag stain technology,we investigated the HBV S gene fragment of sera among 60 cases of children protected by HBV vaccine.Results:in the HBsAg negative sera,there is HBV S gene point mutant which situates 367~369 range of nucleotide from the site of EcoR Ⅰ in the HBV gene group.Conclusions:There is HBV escaping variation in the childern of liver cancer high area.The anti body produced by vaccine stimulation can not neutralize the HBVa diciding cluster having invaded and block HBV infection and it lead to produce 'immune null and void phenomenon'.
