摘要
目的:探讨国人胃癌和结、直肠癌的APC基因突变情况。方法:采用PCR-SSCP银染技术,分析了14例胃癌和14例散发性结、直肠癌的APC基因第15外显子第6段和第7段的突变情况。结果:14份结直肠癌原发灶样品中有1份APC基因第15外显子第7段出现异常电泳条带,而14份胃癌及其6份胃癌转移性淋巴结样品中却未观察到APC基因的异常电泳条带。结论:本组胃癌及淋巴结转移灶中未发现APC基因突变,而结、直肠癌的APC基因突变率为7%。
Objective:To analyse the mutations of the adenomatous polyposis coli(APC) gene in gastric and colorectal carcinomas and thereby to understand the relationship between APC mutation and aggressiveness of gastric and colorectal tumours.Methods:By polymerase chain reaction single-strand conformation polymorphisms(PCR-SSCP) analysis with silver staining,we screened 14 gastric carcinomas and 14colorectal carcinomas for mutations of the exon 15-6 and exon 15-7 segment of APC gene.Results:Mutation was detected in 1 of 14(7%) colorectal carcinomas in the segment 7 of exon 15 of APC gene whereas no abnormal mobility shift was observed in any of other tissue samples such as 14 tumours and 6 lymphatic metastasis of gastric cancer,even though SSCP analysis was extended to the segment 6 and segment 7 of the exon 15.Conclusion:These findings confirm firstly the significance of APC gene mutations of 7% in colorectal,but not gastric carcinomas or its lymphatic metastasis in Chinese.These results may contribute to the shorter segment of APC gene which was analyzed in our study.
