摘要
8310是一种新型M_2受体激动剂,icv 0.5、1、2、4μg/kg呼吸频率、潮气量、每分通气量均明显减少,并呈量效依赖关系,PaO_2明显降低,PaCO_2明显增加;iv 10μg/kg亦致明显呼吸抑制。结果提示8310对呼吸中枢产生抑制,可能与激动呼吸中枢抑制型M_2受体有关。
In the conscious rabbit, the respiratory frequency ( FR ) , tidal volume ( Vt ) , minute ventilation ( Vm ) and the Pa02, PaC02 and PH of the arterial blood were determined. The administration of a novel selective M2 receptor agonist, 8310 0.5-4.0μg/kg icv or 10ng/kg iv, caused decrease in FR, Vt, Vm and PaO2 and increase of PaCO2, and its dose-effect relationship in Vm was linear ( r = 0.998, P<0.01 ) . Our results suggest that the central inhibitory action of 8310 on respiration might be related to the stimulation of the inhibitory M2 cholinergic receptor in respiratory center.
