摘要
本文用中药小檗碱作为探针分子,在0.01mol·L-1醋酸-醋酸钠缓冲体系中,用荧光和紫外-可见吸收光谱研究了贵金属离子Ag?、Au?、Pt?与DNA的相互作用。在三种贵金属离子中,Au?、Pt?离子对小檗碱-DNA体系均具有明显的荧光猝灭作用,而Ag?离子对该体系有强烈的荧光敏化作用。分别求出了三种贵金属离子与DNA的结合常数。三种贵金属离子与DNA结合能力的强弱顺序依次为:Au?>Ag?>Pt?。探讨了三种贵金属离子与DNA的作用机理及导致结合力不同的原因。
The interactions of Ag ??Au ??Pt ?ion with DNA were studied by fluoresc ence spectroscopy and absorption spectroscopy using berberine(BER)as the probe molecule.Obvious spectroscopic changes of the BER-DNA sys-tem in 0.01mol·L -1 HAC-NaAC buffer solution were observed after adding Ag ??Au??Pt ?ion to the binary system,respectively.The appraren t binding constants of these three me tal ions with DNA in the BER-DNA binary system were respectively obtained.According to the binding constants o btained,the order for the binding ab ilities of these three noble metal ions with DNA from the strongest to the weakest was as follows:Au?>Ag ?>Pt ?.Besides,the binding modes of three n oble metal ions to DNA as well as the possible causes of different binding abilities of three noble metal ions with DNA were also discussed in this paper.
