冠心病患者血小板活化及蛋白激酶B的活性变化

摘要 目的 :探讨血小板活化、聚集与PKB/AKT信号通路间的关系 ,研究冠心病发生发展机制。方法 :选取 3 0例UAP、3 2例AMI患者及NC组 3 0人。采用流式细胞仪测定血小板P 选择素表达 ,同时检测血小板PKB放射活性。结果 :UAP和AMI患者血小板P 选择素表达水平均明显高于NC组 (P <0 0 5~ 0 0 1 ) ,但两组间比较无差异 (P >0 0 5 )。UAP和AMI患者血小板胞膜PKB表达均明显高于NC组 (P <0 0 0 1 ) ,而胞浆部分PKB表达均明显低于NC组 (P <0 0 0 1 )。结论 :①与健康对照组相比 ,不稳定心绞痛和急性心肌梗死患者血小板活性明显升高 ;②冠心病患者血小板PKB高活化性与血小板异常活化及血小板血栓形成有关。 Objective: To assess the effect of platelet activation and protein kinase B (PKB/AKT) signal pathway and to study the pathogenesis of coronary heart disease (CHD). Methods: 30 csaes of unstable angina pectoris (UAP) and 32 cases of acute myocardial infarction (AMI) and 30 normal people were selected. Blood samples were drawn 10 min after the treatment. Using flow cytometry technique, the expression of P-selectin were analyzed. In addition, to determine PKB activity of the platelet. Results: Compare with control, P-selectin in UAP patients and AMI patients were higher (P<0.05~0.01). But P-selectin was no differences occurred in the two groups (P>0.05). The PKB in membrane of platelets in UAP patients and AMI patients was higher than in control (P<0.001). The activity of PKB in cytosol of platelets in UAP patients and AMI patients was lower than in control (P<0.01~0.001). Conclusions: ① In UAP patients and AMI patients, the platelet activation was significantly higher. ②The results suggest that the activated activities for platelet PKB was the causes of unusual activated platelet and platelet thrombosis in coronary heart disease.
