应用核酸探针检测水稻细菌条斑菌的研究

摘要 本文报道了一个能有效地同水稻细菌条斑菌的提纯16s rRNA和溶解菌体的粗DNA进行分子杂交的DNA探针.该探针还能成功地检测带菌样本,具有实际应用价值.这个命名为XP_2的探针是12个碱基长度的寡核苷酸探针,其碱基序列是5′ATTGGTAGCCCA.该探针用DNA合成仪合成,经Sephadex G-50层析纯化,在多核苷酸激酶作用下,用〔r-^(32)P〕ATP标记核苷酸5′末端.文中还报道了该探针杂交的条件以及探针的特异性和灵敏度. This paper reports the successful development of a DNA hybridization Probe that performs well with both purified 16s rRNA and crude DNA from lysed bacterial cells of Xanthomonas compenstris pv. oryzicola and has the potential to detect the bacteria from diseased samples. The Probe, named as Xp2, is a oligonucleotide probe and has the sequence 5'ATTGGTAGCCCA. The oligonucleotide probe, 12 bases in length, was synthesized on a DNA synthsizer, Purified by Sephadex G-50 chro-matograph and labled on the 5' end with [r-32P] ATP and Polynucleotide kinase.The conditions are presented for the use of the oligonucleotide probe in RNA and crude DNA slot-blot hybridization.The sensitivity and specificity of the probe were examined.
