摘要
绿秆芋茎尖可在MS+0.2mg/l NAA+4mg/l BA+2mg/lABA+20g/l 甘露醇的培养基上,于15℃、100lx 光照的条件下保存350天,单芽存活率达90%以上,且具良好的生长状态与恢复分化的能力。以芽的叶柄为外植体,在MS+0.2mg/l NAA+4mg/l BA 培养基中培养,一代繁殖系数可达5.5以上。
The shoot tips of Green Stem taro(Colocasia esculenta Schott var.cormosa Chang),a common cultivar in China,were cultured in different conditions.The best result of their invitro preservation was obtained on the medium of“MS+0.2mg/1 NAA+4mg/1BA+2mg/1 ABA+20g/1 Mannitol”at 15℃ and 1000 lux of light.After 350 days of invitro culture,over 90% of the explants survived with satisfactory potential of growth anddifferentiation.Cultured on the medium of“MS+0.2mg/1 NAA+4mg/1 BA”,the petiolcsof buds rctained high propagation coefficient(above 5.5 per generation).
