摘要
用RT PCR法从人胎儿脐静脉内皮细胞中克隆人VEGF受体II基因 3区 ,将其重组到 pAYZ表达载体中 ,构建成人VEGF受体II基因 3区表达载体。将该载体转化大肠杆菌 16C9,获得稳定表达 ,表达产物以可溶性状态存在 ;SDS PAGE和Westernblot分析显示 ,其分子量约为 14kD。以纯化的表达产物免疫Balb/c小鼠 ,眼球采血制备抗血清 ,抗血清效价在 10 3以上 ,并具有抑制VEGF诱导内皮细胞增殖的作用 ,在抑制肿瘤血管新生中具有潜在的应用前景。
RT PCR was used to clone DNA fragment of domain3 of VEGF receptor II (KDRIII ) from human umbilical vein endothelial cell, and the expression vector of KDRIII was constructed by cloning KDRIII DNA fragment into the expression vector pAYZ KDRIII was expressed in E coli 16C9 and purified by affinity chromatography The serum against KDRIII was prepared from mice immunized with a soluble form of KDRIII The serum against KDRIII suppressed proliferation of ECV307 induced by VEGF and have potential clinical application in the treatment of cancer where pathological angiogenesis is involved
