Enzymatic Synthesis of Biosurfactant-fatty Acid Glycoside Esters

摘要 以固定化脂肪酶作生物催化剂在无溶剂体系中成功地合成了乙基葡萄糖苷油酸单酯。系统地研究了不同来源的酶、糖苷种类、初始水分、温度和酶的浓度等因素对酶催化反应的影响。生物催化剂 Novozym435可循环使用 8次以上且无明显酶活损失。确定出最适的反应条件为 :温度80°C,初始水分为 0 ,酶的浓度为 w酶 =0 .0 5 ,反应时间为 1 2 h,油酸的转化率 1 0 0 % ,单酯得率为86.5 5 % Oleic acid ethyl glycoside esters were synthesized by enzymatic esterification of ethyl glycoside in solvent free system. Operational conditions were first optimized using oleic acid as acyl group. Several factors, such as lipase origin, temperature, water content and enzyme concentration, have been studied systematically in the esterification reaction. The operational conditions have been optimized: 200 r/min, 80°C , initial water content 0, enzyme concentration 5% and time 12h. The biocatalyst Novozym 435 could be recycled more than eight times without any significant activity loss.
