摘要
目的本文探讨雌二醇对去卵巢后高血压大鼠心血管系统的作用。方法 1 2周龄雌性自发性高血压大鼠 1 8只 ,随机分成 3组 :实验组 2组 ,大鼠去卵巢后分别喂养安慰剂、雌二醇 1mg/kg/day ;未去卵巢组 1组给安慰剂喂养作为对照组。分别测量治疗 3个月前后收缩期血压、体重、全心重量、左心室重量、血浆AngⅡ浓度、血脂、血糖和尿素氮肌酐、尿酸以及肠系膜三级动脉血管壁 /腔面积比。结果 (1 )与未去卵巢组比 ,去卵巢后收缩期血压明显升高。雌二醇组治疗后收缩期血压均明显下降 ;(2 )去卵巢后全心重量和体重之比及左心室重量和体重之比明显高于未去卵巢组 ;(3)去卵巢后血浆AngⅡ浓度明显升高 ,雌二醇治疗后AngⅡ浓度明显下降 ;(4)去卵巢后血浆TC、TG明显升高 ,雌二醇治疗后TC、TG浓度下降 ,去卵巢后血糖明显升高 ,治疗后降低 ;(5)去卵巢安慰剂组肠系膜动脉壁 /腔面积比明显高于未去卵巢组 ,去卵巢雌激素喂养组比去卵巢安慰剂组明显下降。结论 去卵巢对心血管系统具有不利影响。雌激素替代治疗消除或减少不利影响 。
Objective To investigate the cardiovaso protective effects by estradiol treatment in the ovariectomied spontaneously hypertensive rats. Methods Eighteen female spontaneously hypertension rats were randomly divided into 3 groups. In two groups, rats were treated with placebo or estradiol (1 mg/kg/day), after ovariectomy, with another group without ovariectomy served as control. Rats were fed with chow diet. The systolic blood pressure, body weigh, heart weigh, weigh for left ventricle of heart, plasma concentration of angiotensin Ⅱ, lipid, glucose, urea nitrogen, creatinine, uric acid, and mesenteric arteries (3rd grade branch) wall to lumen ratio were determined 3 months after the treatment. Results 1.The systolic blood pressure (SBP) was significantly increased in ovariectomied rats . The SBP was significantly decreased in ovariectomied rats when they were treated with estradiol. 2.The heart to body weigh ratio and left ventricle of heart to body weigh ratio in ovariectomied rats were significantly higher than that in non ovariectomied rats. Both ratios were significantly decreased after treatment with estradiol; 3.Plasma concentration of angiotensin Ⅱ was significantly increased after ovariectomy. Estradiol treatment significantly decreased plasma Ang Ⅱ. 4.Plasma glucose, cholesterol, triglyceride and high density lipoprotein were increased after ovariectomy. Estradiol treatment decreased plasma cholesterol and glucose. 5.Histological examination showed that W/L of mesenteric arteries (3rd grade branch) was significantly lower in ovariectomied rats than in non ovariectomied rats. The ratios were significantly decreased after treatment with estradiol. Conclusion Wide variety of adverse effects on cardiovascular system were shown after ovariectomy in rats. Estrogen replacement therapy eliminated or reduced the adverse effects. Which was potentially benefietal to the protection cardiovascular system.
