兔角膜重度碱烧伤后羊膜移植的疗效观察 被引量:8

摘要 目的 观察兔角膜重度碱烧伤后羊膜移植的疗效。方法 将 18只新西兰白兔随机分成 3组 ,每组 6只 ,所有兔眼建立重度碱烧伤模型 ,左眼羊膜移植 ,右眼作为对照。分别于 1、2、3个月 ,通过光镜、电镜以及原位杂交方法观察角膜情况。结果  1个月时 ,羊膜移植组上皮细胞修复优于对照组 ,基质浸润轻于对照组 ,基质中 TGF- β1 m RNA的表达低于对照组 ,而新生血管增生情况两者无差别。2个月时 ,除上皮细胞修复羊膜移植组优于对照组外 ,其他与对照组相比无差异。3个月组情况与 2个月组相似。结论 羊膜移植治疗兔角膜重度碱烧伤 ,早期可促进角膜上皮细胞修复 。 Objective To observe the therapeutic effects of amniotic membrane transplantation in severe alkali burn of rabbits' corneas.Methods Eighteen New Zealand white rabbits were divided into three groups randomly,each having 6 rabbits. The models of severe corneal alkali burns were set up in all rabbits, then left eyes were transplanted with amniotic membrane (AM group) and right eyes were served as control(C group). After 1 month, 2 months, 3 months, the corneal conditions were observed by light microscope, transmission electron microscope, in situ hybridization.Results At the first month, it was found corneal epithelia reconstructed better in AM group than that in C group and stromal infiltration was less than that in C group. At the same time, it was found that the expression of TGF-β 1 mRNA in AM group was less than that in C group, but neovascularization was alike in two groups. At the second month, corneal epithelia reconstruction was also better in AM group, but others were alike in two groups. At the third month,the condition was the same as that at the second month.Conclusion In early period, for severe corneal alkali burns, the transplantation of amniotic membrane is an effective method for reconstructing the corneal epithelial defect and reducing stromal infiltration.
