摘要
目的 :研究 p5 3和 bcl-2的基因产物在正常口腔粘膜 (NOM)、口腔粘膜白斑 (OLK)及口腔鳞癌 (OSCC)中的表达及意义。方法 :应用免疫组织化学 S-P法 ,对 NOM(1 0例 )、OLK(1 7例 )、OSCC(3 2例 )中 p5 3和 bcl-2的表达情况进行测定。结果 :p5 3在正常 NOM中无表达 ,在 OLK中有中度表达 (阳性率 2 9.4% ) ,在 OSCC中有较强的表达 (阳性率 5 6.2 5 % ) ;bcl-2在 NOM中无表达 ,在 OLK中有较强的表达 (阳性率 2 3 .5 3 % ) ,而在 OSCC中的表达 (阳性率 2 1 .88% )与 OLK中的表达比较差异无显著性。结论 :1 p5 3的突变和 bcl-2的过度表达 ,通过抑制细胞凋亡促进了OLK和 OSCC的形成 ;2在 OLK的形成过程中 ,突变型 p5 3和 bcl-2的基因产物表达呈正相关 ,而在 OSCC的形成过程中 ,二者的基因产物无明显相关性 ;3 bcl-2在 OLK和 OSCC的形成过程中 ,并非单纯通过抑制细胞凋亡这一机制而起作用 ;4突变型 p5 3的表达可作为 OLK及 OSCC的基因标志物。
Objective:To investigate the expression of p53 and bcl 2 gene producs in normal oral mucosa, oral leukoplakia and oral squamous cell carcinoma and to study reationship between both expressions of p53 and bcl 2 protein and the development of OLK and OSCC. Methods: Immunohistochemical S P method was used in the experiment to detect the bcl 2 and p53 protein expressions in 10 normal oral mucosae, 17 oral leukoplakeas and 32 squamous cell carcinoma.Results:There was no expression of p53 protein in normal oral mucosa. But moderately positive expression of p53 protein in OLK and strongly positive expression of p53 protein was found in OSCC; bcl 2 protein expression was observed in basal cells of normal oral mucosa, moderately positive expression of bcl 2 protein was detected in OLK and OSCC, and there was no significant difference between both of them.Conclusion:① The mutation of p53 or overexpression of bcl 2 may play an imprtant role in the development of OLK and OSCC by preventing apoptosis. ② There is a positive relationship between expression of p53 mutation and bcl 2 in the development of OLK but no corresponding relationship between both of them in the development of OSCC. ③ In the development of OLK and OSCC, bcl 2 plays its role not only by preventing apoptosis. ④ The expression of mutated p53 could be regarded as the genetic marker of OLK and OSCC.
