摘要
目的研究雄激素和雌激素对培养的人前列腺间质细胞碱性成纤维细胞生长因子(bFGF)和转化生长因子(TGFβ2)表达的影响。方法本实验培养出11株良性前列腺增生症(BPH)病人前列腺间质细胞,并给予双氢睾酮(DHT)和雌二醇(E2)刺激,通过RT-PCR法观察bFGF和TGFβ2的mRNA表达,同时观察了平滑肌细胞分化特异性抗原(smoothelin)的mRNA表达。结果DHT能明显上调bFGF表达,E2能明显上调TGFβ2和smoothelin表达,且TGFβ2和smoothelin表达量成正相关。结论DHT能诱导bFGF表达,E2能诱导TGFβ2表达。E2诱导向平滑肌的细胞转化可能与TGFβ2有关。
Objective To studyeffectsof androgenandestrogenon theexpressionsof basicfibroblastgrowthfactor(bFGF)andtransforminggrowthfactorβ2(TGFβ2)inculturedhumanprostaticstromalcells.Methods Humanprostaticstromal cellsobtainedfrom11patientswithbenignprostatichypertrophy(BPH)wereculturedandstimulatedwithdihydrotesto-sterone(DHT)andestradiol(E 2 ).Theexpressionsof bFGFandTGFβ2mRNAalongwithsmoothelinmRNAwereobservedwith reversetranscriptase-polymerasechainreaction.Results DHT significantlyupregulatedbFGFexpression,andE 2 enhanced TGFβ2andsmoothelinexpressions.A positivecorrelationbetweenexpressionsof TGFβ2andsmoothelinwas observed.Conclusion DHTcaninducebFGFexpressionandE 2 promotes TGFβ2expression,andthetransformationtowardsmooth musclecellsinducedby E 2 mayinvolvetheactionof TGFβ2.
