氧氟沙星对人体内茶碱稳态血浓度的影响 被引量:2

The Influence of Ofloxacin on Human Serum Theophylline Concentration in Steady State

摘要 目的:探讨氧氟沙星对人体内茶碱稳态血浓是否存在影响,以便指导临床医师合理用药。方法:24例呼吸系统疾病的入院患者,口服氨茶碱片剂,0.1g,tid(8:OO,12: 00和16:OO时),加静滴氨茶碱0.25g,同时口服氧氟沙星片,0.2g,bid(8:00和20:00时)。第5d早晨服药前取静脉血测定联合用药后茶碱最低稳态血浓。当日起停服氧氟沙星,氨茶碱用量不变,至第8d再行测定茶碱最低稳态血浓。结果:合用氧氟沙星后茶碱平均稳态谷浓度为12.08±3.76mg·L-1,茶碱单用时平均稳态谷浓度为11.39±3.15mg·L-1,二者无显著性差异(P>0.05)。结论:氧氟沙星对茶碱在人体中的稳态血浓无明显影响,临床将二者合用时毋需事先减少茶碱剂量。 OBJECTIVE: To investigate whether ofloxacin may influence theophyllineserum concentration in human bodies and whether it's necessary to adjust the dosageof theophylline in advance when the two drugs were administered concurrently, so asto conduct clinicians to prescribe ratonally. Methods: 24 inpatients contracted chronicbronchitis, bronchial asthma or pneumonia had been monitored and evaluated in thestudy. Ofloxacin capsules (0.2g) were orally given every l2h (8.00 and 20:00 ) andaminophylline tablets (0.lg) were given three times daily (8:00, l2:00, l6:00) with aadditional intravenous infusion of aminophylline (0.25g diluted in glucose solution)at 20o'clock to each patient for four days. Ofloxacin capsules were withdrew from the5th day and administrations of aminophylline were continued. The minimal steady-state serum concentrations of theophylline were determined on the morning of 5th andof 8th day before the administration. RESULTS: It was found that the average minimalconcentration of serum theophylline in steady state was l2.08±3.76 mg.L-1 withconcomitant ofloxacin, and ll.39±3.l5 mg.L-1 without ofloxacin.There were nostatistical significance between theophylline concentraion with and without ofloxacin(P>0.05). CONCLUSIONS: Ofloxacin had minor influence on serum theophyllineconcentration in human bodies and it demonstrated that theophylline dosageadjustment did not appear to be necessary in patients receiving concomitant ofloxacinin clinics.
