摘要
目的 探讨细胞周期调节蛋白 p16、cyclin D1和 Rb在宫颈上皮癌变过程中的表达及相互作用。方法 应用免疫组化技术 (S- P法 ) ,检测 p16、cyclin D1和 Rb基因在 10例正常宫颈组织、10例慢性宫颈炎、10例宫颈上皮内瘤样变 (CIN)及 5 9例宫颈鳞癌中的表达。结果 10例 CIN中 2例 p16蛋白阴性表达 ,占 2 0 .0 % ,5 9例宫颈鳞癌中 ,17例 p16免疫反应阴性 ,占 2 8.8% ,两者之间差异无显著性 (P>0 .0 5 )。宫颈上皮癌变过程中 (慢性宫颈炎→上皮内瘤样变→浸润癌 ) cyclin D1过表达率逐级升高 (10 .0 % ,40 .0 % ,74.6 % )。宫颈癌中 Rb表达阳性率为 88.1%(5 2 / 5 9)。结论 p16失活参与部分宫颈癌的发生。 cyclin D1表达异常与宫颈癌发生有关 ,其过表达出现在宫颈上皮癌变的早期阶段。 Rb可能主要通过功能性失活参与宫颈癌的发生。
Objective To study the expression of p16,cyclin D1 and Rb in the carcinogenesis of cervix. Methods Using S-P immunohistochemical technique,we examined the expression of p16,cyclin D1 and Rb in 10 cases of normal cervical tissues,10 chronic cervicitis,10 cervical intraepithelial neoplasias (CIN) and 59 invasive squamous cell carcinoma (CIN). Results 20.0% (2/10) CIN and 28.8% cervical carcinomas (17/59) showed negative p16 staining ( P >0.05). Rb was expressed in 52 of 59 (88.1%) cases of cervical carcinomas. In cervical carcinogenesis,with the lesions progressing,the overexpression rate of cyclin D1 protein had a increasing tendency (10.0%→40.0%→74.6%). Conclusion Inactivation of p16 may be involved in the genesis of human cervical carcinoma. Rb may be functionally inactivated in cervical cancer. Overexpression of cyclin D1 may have a significant role in the pathogensis of invasive cervical squamous carcinoma.\;
