摘要
为探讨宫内急性缺血缺氧及再灌注时细胞间粘附分子 1(ICAM 1)在胎鼠肾脏损伤发生中的作用 ,通过钳夹孕 2 1日龄大鼠供应子宫的动静脉血管以制备胎鼠宫内不同程度缺血缺氧模型和再灌注不同时间模型 ;利用逆转录聚合酶链式反应动态观察ICAM 1mRNA的变化。结果显示 :正常时ICAM 1mRNA在 2 1日龄胎鼠肾脏有一定表达。宫内缺血后 ,胎肾ICAM 1mRNA在缺血 35分钟时开始表达增强 (P <0 0 5 ) ,4 5分钟时达高峰 (P <0 0 1)。缺血 15分钟再灌注过程中 ,胎肾ICAM 1mRNA在再灌注 1小时时即开始表达增强 (P <0 0 5 ) ,8和 15小时时为表达高峰 ,明显强于其它各时间点 (P <0 0 5 ) ,此后表达下降 ,在 30小时已接近假手术组水平 (P >0 0 5 )。因此 ,可以认为宫内缺血缺氧可以刺激胎鼠肾脏ICAM 1mRNA的转录 ,提示ICAM 1可能和窒息后肾损伤发病关系密切。
To study the effect of intercellular adhesion molecule 1(ICAM 1)on renal injury after intrauterine acute ischemia,we made the models of intrauterine ischemia and reperfusion in different times by clamping one side of vessels supplying blood to uterus of Wister rats of 21 gestational days. The dynamic changes of ICAM 1 mRNA in the fetal kidney were detected by the method of reverse transcription polymerse chain reaction.The results showed that there was a certain quantity expression of ICAM 1 mRNA in the sham operation group.The expression of ICAM 1 mRNA began to increase on 35 minutes after ischemia ( P <0.05) and its peaking time was 45 minutes after ischemia( P <0.01).During reperfused stage after ischemia for 15 minutes,it has begun to rise at reperfused 1 hour( P <0.05).The highest period was in reperfused 8 to 15 hours( P <0.01).It recovered at reperfused 30 hour( P >0 05). In conclusin, ischemia and reperfusion can induce the elevation of ICAM 1 mRNA. It suggest ICAM 1 play an important role in the development of asphyxia induced renal injury.
