SiO_2对巨噬细胞钙稳态的影响

摘要 使用荧光染料Indo-1测定法探讨了SiO_2对肺泡巨噬细胞胞浆游离钙水平的影响。结果表明,SiO_2对巨噬细胞胞浆游离钙水平的升高有明显的剂量效应关系。随着SiO_2作用时间的延长,胞浆游离钙水平呈持续性升高,并与培养液中LDH活性上升相平行。给予柠檬酸铝预处理的SiO_2可防止胞浆游离钙的增加。提示,SiO_2可引起巨噬细胞内钙稳态紊乱,这一紊乱与SiO_2对细胞膜的损伤有密切联系。 The effect of silica on cytoplasmic calcium concentration of macrophages was studied with fluo-resent calcium indicator indo-1. The results showed that an evident dose-effect relationship exists between the rise in cytoplasmic free calcium and silica doses in the process of silica-induce macro-phage damage. Apart from instant rise in cytoplasmic free calcium by stimulating receptor,the level of cytoplasmic free calcium of macrophages were characterized by continuing rise with longer period of the interaction between silica and macrophages,and the rise in cytoplasmic free calcium were parallel to the increase of LDH activity in culture medium. In contrast, silica pretreated by aluminium citrate could prevent from the rise in cytoplasmic free calcium. All those results indicated that silica can cause the disarrangement of calcium homeostasis of macrophages and the disarrangement has a close relation to silica-induced membrane injury.
