摘要
采用顶空GC/MS多离子检测技术和毒物动力学分析方法,对暴露于400ppm苯、乙苯和二甲苯中的家兔,定时观测血液毒物浓度(C_b)的时间过程。在暴露6小时期间内,C_b迅速上升,并分别维持在坪浓度2.05~4.46μg/ml处。在暴露终止后,C_b呈双相半对数-线性衰减,与二室线性毒物动力学模型计算的浓度-时间曲线吻合。
Benzene, ethylbenzene and xylene are common aromatic hydrocarbon compounds. Their toxicoki--netics were studied in 15 healthy rabbits, which were exposed for 6 h to 400 ppm of the compoun ds, respectively. Head space GC/MS MID technique was used to measure their concentrations in blood at regular times for observing time course of the compounds. Concentrations of the compounds plotted against time stowed that it rose quickly and maintained a plateau of approximately 2.05-4.46μg/ml throughout 6 h exposure period and that it exhibited a biphasic log-linear decline after the exposure. The observed conentrations of the compounds in blood were fit to the concentrations vs. time curve predicted by the twocompartment linear pharmacokinetic model. The half-life values obtained for the rapid (α) and slower (β) elimination Phases were 0.11-0.20h and 1.5-2.5 h, respectively. The studies indicated that benzene and ethylbenzene were of similar toxicokinetic characteristices and were of more concentrations in blood than xylene.
