摘要
在分离并系统鉴定了减蛋综合症病毒AA_2株的基础上 ,进行了该毒株的体外培养试验 ,以寻求适应于细胞培养物中继代的细胞适应毒。结果表明 ,AA_2毒株可在鸭胚成纤维细胞 (DEF)、鸭胚肾细胞 (DEK)、鸭胚肝细胞 (DEL)、鸡胚成纤维细胞 (CEF)、鸡胚肾细胞 (CEK)和鸡胚肝细胞 (CEL)上增殖和继代 ,并随着继代次数的增加 ,CPE潜伏期缩短 ,持续期延长 ,半数细胞感染量 (TCID50 )呈逐代增加趋势 ,血凝效价 (HA)逐代稳步增强 ,至第 15代之后 ,这些变化趋于稳定。证明用鸭胚细胞培养的AA_2株CPE优于鸡胚的 ,在肝细胞培养物收获液中 ,其单位体积中的TCID50 和HA效价略高于肾细胞和成纤维细胞。末发现AA_2株病毒在PK_15 ,BHK_2 1,SP2 / 0等细胞中增殖的迹象。还较为系统的测定了第 19代DEF毒的其它特性。发现在接毒后 12 0小时 ,HA及TCID50 值最高 ,细胞适应毒对鸭胚致死率为 10 % ,比原毒降低 2 3% ,理化特性与原毒一致 。
Egg drop syndrome virus(EDSV)is blongs to the Aviadenovirus group Ⅲ according to its phisi-chemical and imnurological.Here we discribed the characterization of a Chinese isolate strain AA2 of EDSV in various cells.It has been shown that the virus multiplied and propagated in cells such as Duck Embryo Fibrocyte(DEF),Duck Embryo Kidney(DEK),Duck Embryo Liver(DEL),Chick Embryo Fibrocyte(CEF),Chick Embryo Kidney(CEK)and Chick Embryo Liver (CEL).As the passages increased,the CPE lasted longer with longer latency.TCID 50 and HA increased steadily with each passage until the 15 th generation,when they reached plateau.It was proved that Duck Embryo (DE) cells overmatched Chick Embryo (CE) cells in CPE by AA2 strain.The titers of TCID 50 and HA from liver cell culture were higher than those with kidney cells or fibroblasts,although the later culture was easily available in large quantities.The present study did not find any evidence of multiplication of AA2 strain in PK-15,BHK-21 and SP2/0.The study also observed characteristics of virus at 19 th generation in fibroblasts.The titers of virus and TCID 50 were highest(5.5 lg and 1∶4096 respectively)when CPE appears as 100%.Inoculation in DE with virus adopted for cells can cause slight lesions with 10% of death,23% lower than that of wild virus.The adaptive virus shared similar chemical and physical characteristics with the wild virus,and its coagulation activities could be repressed by standard EDS virus positive serum,respectively.
