抗金葡萄球菌免疫核酸诱生细胞毒因子的研究

摘要 用抗金黄色葡萄球菌免疫核酸致敏家兔,发现兔血清中含有直接杀伤L929细胞的细胞毒因子。就其成分进行了探讨,推测其中主要成分为肿瘤坏死因子(TNF)。并可有淋巴毒素及起协同作用的少量干扰素等其他一些成分。分别用体外细胞毒实验即结晶紫染色法及地~125I—UDR掺入法,计算细胞毒指数,评估结果,对两种检测手段进行了比较,染色法虽不如同位素法灵敏度高,但无放射性污染、不易保存等缺点。应用放线菌酮可以使细胞毒作用放大,便于检测到较微弱的细胞毒活性,而且实验条件易于控制,结果也较稳定。 In this experiment, the cytotoxic factors were found in the serum from the rabbits sensitized by Sa-iRNA. The factors could directly destroy L929 cells. The elements of the cytotoxic factors were studid. It was supposed that its major elements were TNF, possibly a little lymphotoxin and little IFN. Using the cytotoxic tests, in vitro, the cytotoxic index was caculated, In Comparison with photometric assay isotopic assay was more sensitive、less stable, easily caused radiative contamination and is difficult to restore, Although photometric assay is less sensitive, the cytotoxic activity can be amplified by using Actidione, the weak cytotoxic activity can be detected, More reasonably the experiment conditions are easily controlled, the results were more stable. So it is a simple convenient assay of detecting the cytotoxic activity.
