摘要
目的:从自由基代谢和肿瘤坏死因子(TNF)的变化方面研究益元活血丹和大黄及血栓心脉宁对老龄大鼠脑缺血再灌注胃肠损伤防护作用的机制。方法:老龄(20月龄以上)大鼠分为对照组、模型组、尼莫地平组、益元活血丹大小剂量组、大黄组、血栓心脉宁组,观察大鼠全脑缺血30min再灌注60min后胃肠组织超氧化物歧化酶(SOD)活性和丙二醛(MDA)、TNF含量变化。结果:老龄模型组胃组织MDA/SOD比值高于老龄对照组,而小肠组织的比值有增高趋势,小肠组织中TNF增高;与老龄模型组比较,用药各组胃组织和血栓心脉宁组小肠组织MDA/SOD比值及益元活血丹大小剂量组TNF含量均降低。结论:大黄和益元活血丹及血栓心脉宁对脑缺血再灌注胃肠损伤保护作用机制可能与其拮抗自由基和TNF对组织损伤有关。
To study the guarding action mechanism of Yiyuanhuoxue pills, Rheubarb and Xueshuanxinmaining capsules on the refilling gastrointestinal hurt of brain-ischemic aged rats from the free-base metabolism and the change of TNF amount. Aged rats (over 20 months old) were divided into the normal control group, Nimotip-ine group,^ major and minor dose Yiyuanhuoxue pills group, Rheubarb group and Xueshuanxinmaining group to observe the SOD activity of gastrointestinal tissue and the amount change of MDA and TNF after the total brain ischemia 30 minutes and refilling 60 minutes later. The results showed that the ratio of MDA/SOD of gastrie tissue in aged model group was higher than the aged control group; the ratio of enteric tissue tended to increase; TNF increased in enteric tissue; the ratio of MDA/SOD of gastric tissue in medication groups and intestinal tissue in Xueshuanxinmaining group decreased compared with the aged model group, so did the amount of TNF in major and minor dose group of Yiyuanhuoxue pills group. Thus the guarding mechanism from gastrointestinal hurt of brain-ischemic refilling of Rheubarb, Yiyuanhuoxue pills and Xueshuanxinmaining capsules was likely to have relation with their anti-free-base and preventing TNF from hurting the tissue.
