本文报道对 Spitz 等方法略加改进后,以 SMMC-7721人体肝癌细胞系的完整细胞和牛甲状腺球蛋白(BTG)为免疫原,经小鼠脾内一次免疫后观察到的体液免疫应答结果.
This paper discribes the possibility of applying a modified primary intrasplenic immunization for preparation of monoclonal antibodies.The SMMC-7721 cell line,a human liver carcinoma cell line,and bovine thyroglobulin(BTG)were used for immunization of BALB/C mice with this procedure.The titers of serum antibodies of the immunized mice were tested by indirect immunofluorescence assay and compared with those after an intraperitoneal injection of the antigens.We found that a small dose of 3×10~5cells or 20μg protein in the modi- fied primary intrasplenic immunization is as effective to elicit humoral immune respose as a large dose of 1×10~7cells in conventional intraperitoneal immunization.Therefore it is reasona- ble that the modified primary intrasplenic immunization can be used for production of monoclinal antobpdoes.
