摘要
目的 :观察成肌细胞自体移植后在宿主体内生长、增殖情况及对实验动物心功能的影响。方法 :将 30只SD大鼠随机分为移植组 (n =2 0 )及对照组 (n =10 ) ,另选 6只进行皮下自体移植 ,液氮冷冻大鼠左心室游离壁 ,建立心肌梗死模型。将体外培养的成年大鼠成肌细胞自体移植到相应大鼠的皮下及心肌疤痕中 ,观察细胞的生长、增殖情况 ,结合心脏重量指数及血流动力学等参数评价细胞移植后对心功能的影响。结果 :6只进行皮下自体移植的大鼠中 ,有 2只在移植部位形成排列紊乱的肌纤维结构。 2 0只行成肌细胞心肌内自体移植的大鼠中 ,有 6只myogenin染色阳性 ,证明成肌细胞可在宿主皮下及心肌疤痕中存活。与移植失败者及对照组相比 ,移植成功者左心室重量、左心室重量 /体重、心脏重量 /体重及左心室舒张末期压力均降低 ,有统计学差异 (P <0 0 5~ 0 0 1)。压力变化率最大值 (dp/dtmax)升高 ,有统计学差异 (P <0 0 5 )。结论 :成肌细胞自体移植后可在宿主体内生长、增殖 。
Objective:This study was to investigate proliferation and functions of in the hosts after auto-transplantion. Methods:Approximately 7×10 6 skeletal myoblasts were cultivated in vitro from adult Sprague-Dawley rats whose cardiac apexes had been cryoinjured to create a transmural lesion.Two weeks after cryoinjury,autologous myoblasts or serum-free culture medium was injectedinto the scar tissue of transplantation (n=20) and control (n=10) animals,respectively.Four weeks after injection,heart function into the two groups was measured using a MPA-V instrument.Hearts were harvested and histologic studies were performed to quantify the transplanted cells.Also almost 7×10 6 skeletal myoblasts were auto-transplanted in subcutaneous connective tissue of the rats' hindlimbs (n=6).The other hindlimbs were injected with 0.25ml serum-free Ham's F-10 medium. Results:After cryolesion,histologically,the transmural lesion contained inflammatory cells and active scarring but no viable cardiomyocytes.Six hearts contained myocytes in the cryolesion,and immunohistochemical analysis confirmed their skeletal muscle origin.Multinucleated myocytes were also found in the adult rats' subcutaneous connective tissue of two hindlimbs.Compared with the control group and failure group,successful transplantation group had lower left ventricle weight(LVW,p<0.05),LVW/BW(p<0.05),HW/BW(p<0.05),and left ventricular end-diastolic pressure(LVEDP,p<0.01)and higher dp/dt max(p<0.05). Conclusion:Skeletal myoblasts can survive in the hosts' scar tissue.Heart function can be improved after auto-transplantation.These experiments set the stage for future studies on heart function of patients with severe ischaemic heart failure.
