转化生长因子β、癌基因c-myc和c-fos在瘢痕形成中的作用 被引量:10

摘要 目的了解c-myc和c-fos蛋白产物在瘢痕中的表达情况以及转化生长因子β(TGF-β1)对其表达及胶原合成分泌的影响,探讨瘢痕增生机制。方法采用免疫组织化学方法对增生性瘢痕(HS)、非增生性瘢痕(NHS)和正常皮肤各9例标本进行了c-myc和c-fos蛋白产物的检测和比较;通过细胞培养的方法研究了TGF-β1对增生性瘢痕成纤维细胞癌基因c-myc和c-fos的表达及胶原分泌的影响。结果癌基因c-myc和c-fos蛋白产物在增生性瘢痕成纤维细胞中呈强阳性表达,TGF-β1可显著提高HS成纤维细胞c-myc和c-fos的表达及胶原合成分泌的增加。结论c-myc和c-fos蛋白产物可能是TGF-β1引起成纤维细胞内生物学效应的中间信号途径。 Objective To investigate the localization of c myc and c fos products in hypertrophic scar, the effects of TGF β1 on expression of them and on synthesis of collage of fibroblast in hypertrophic scar. Method Respective 9 specimens of HS,NHS and skin tissues were stained with immunohistochemical method for c myc and c fos products.The means of cell culture was used to investigate the effects of TGF β1 on expression of oncogene c myc and c fos and on synthesis of collage. Results The result demonstrated significant numbers of fibroblasts were positively stained for c myc and c fos,TGF β1 can significantly increased the expression of c myc and c fos and increared 2 times the synthesis of collagen of fibroblast cell in hypertrophic scar.Conclusion c myc and c fos protein products may localize along an axis of TGF β receptor mediated intracellular signaling pathway.
