摘要
目的 观察日本血吸虫 (中国大陆株 )的线粒体相关蛋白 r Sj338及膜蛋白 r Sj2 2 .6有效抗原表位混合后对小鼠的免疫保护性 ,为将来复合疫苗的研制提供理论依据。方法 分别用特异性抗r Sj338抗体和抗 r Sj2 2 .6的抗体筛选噬菌体随机 12肽库 ,将获得的 r Sj338的有效抗原表位与r Sj2 2 .6有效抗表位混合后免疫 BAL B/ c小鼠 ,并与 r Sj338的有效抗原表位混合免疫组进行比较。结果 r Sj338的 4种表位和 r Sj2 2 .6抗原的 4种表位混合免疫组小鼠获得了 4 5 .4 % (P<0 .0 1)的减虫率及 5 9.1% (P<0 .0 1)肝总减卵率 ,r Sj338的 4种表位混合免疫组小鼠获得了 34.2 % (P<0 .0 1)的减虫率及 4 6 .8% (P<0 .0 1)肝总减卵率 ;r Sj3384种表位和 r Sj2 2 .6的 4种表位混合免疫组获得的减虫率和肝总减卵率均高于 r Sj338的 4种表位混合免疫组 ,差异有显著性 (P<0 .0 5 )。
Objective To evaluate the immunoprotective effect of the mixture of four rSj338 epitopes and four rSj22 6 epitopes of Schistosoma japonicum. Methods A 12 mer peptide phage library was used to screen the epitopes of rSj338 and rSj22 6 with the purified polyclonal IgG antibody against rSj338 and rSj22 6, respectively. The mice were immunized with the mixture of the epitopes and challeged with S.japonicum cercariae. Results The mice immunized with the mixture of four rSj338 epitopes and four rSj22 6 epitopes induced 45 4% (P<0 01) worm redcution and 59 1% (P<0 01) total liver egg reduction. The mixture of only four rSj338 epitopes induced 34 2% (P<0 01) worm reduction and 46 8%(P<0 01) total liver egg reduction. The reduction rates in former mixture were higher than those in latter mixture (P<0 01). Conclusion The immunoprotective effect of the mixtue of rSj338 epitopes and rSj22 6 epitopes was superior to the result obtained from mice immunized with the mixture of only four rSj338 epitopes.
