摘要
目的:对重组Sj22.6kDa抗原(rSj22.6)进行免疫学活性鉴定,了解其作为疫苗候选抗原的潜力。方法:Westernblot反应确定rSj22.6的抗原性;将rSj22.6注射家兔,制备特异抗血清;以血吸虫尾蚴对C57小鼠进行攻击感染,初步鉴定其免疫保护力。结果:rSj22.6可与特异性抗体发生反应,并刺激家兔产生特异性抗体应答,抗体滴度为11280。攻击感染中,免疫组小鼠的减虫率达77.2%。结论:rSj22.6具有良好的免疫学活性,在诱导抗日本血吸虫感染方面具有一定的潜力。
AIM:To identify the immunogenicity and the potentiality of r Sj 22.6 as a candidate vaccine antigen for schistosomiasis japonica. METHODS:r Sj 22.6 was tested by Western blot with specific antibodies to identify its immunogenicity.The r Sj 22.6 protein was isolated and purified by SDS PAGE and injected twice into rabbits to raise antir Sj 22.6 serum that was tested by Western blot to recognize the native protein of adult S.japonicum and SEA.C57 mice were immunized with r Sj 22.6 and challenged with S.japonicum cercariae to identify the protective immunity raised by r Sj 22.6.RESULTS: r Sj 22.6 could be recognized by specific antibodies and could stimulate the rabbits to produce high level of anti r Sj 22.6 antibodies. Anti r Sj 22.6 serum could recognize an adult native protein with the same molecular weight as r Sj 22.6.The preliminary immune challenge test resulted in significant reduction in adult worm burden. CONCLUSION: r Sj 22.6 possesses immunogenicity in stimulating rabbits to produce high level of specific antibody and has protective role as a candidate vaccine antigen against S.japonicum infection.
