摘要
目的 :探讨临床应用异种无细胞真皮基质 (Xeno ADM)的免疫反应机理。方法 :分离正常猪、正常人和接受Xeno -ADM复合移植的临床大面积烧伤患者的外周血单个核细胞 (PBMC) ,以前两种细胞为刺激细胞、后者为反应细胞 ,通过异种混合淋巴细胞培养 (Xeno -MLC)和同种异体混合淋巴细胞培养 (Allo -MLC)、3 H TdR掺入和酶联免疫吸附分析 (ELISA)方法 ,检测不同抗原对反应细胞增殖的刺激作用及其IL - 2 (IU·ml-1)、IL - 4 (IU·ml-1)和IFN - (ng·L-1)的动态变化。结果 :Xeno MLC组淋巴细胞增殖反应和培养 4d后的IL - 2、IL - 4、IFN - 的分泌水平均明显高于Allo -MLC组和空白对照组 (P <0 .0 5~ 0 .0 1)。结论 :有辅助性T淋巴细胞 - 1(Th1)和Th2参与的细胞免疫和IL - 4诱导的体液免疫可能是异种移植物 (Xeno -ADM)免疫排异反应的特征 ;用预先致敏的淋巴细胞进行MLC 。
Objective:To investigate the immune response mechanism involved in clinical transplantion of xenogeneic acellular dermal matrix(Xeno-ADM).Methods:Peripheric blood mononuclear cells(PBMC)were separated respectively from normal pigs and humans,and patients suffered extensive burn accepted the transplantation of Xeno-ADM.Then the mixed lymphocyte cultrue(MLC)was performed through co culturing PBMC of the patients with PBMC of the normal pigs or humans.The cells proliferation,and secretion of IL-2(IU·ml -1 ),IL-4(IU·ml -1 )and IFN-(ng·L -1 )were measured by 3 H-TdR incorporation and enzyme linked immunosorbent assay(ELISA).Results:At 4 days after culture,the cells proliferation and secretion of IL-2,IL-4,IFN were significantly higher in MLC of normal pigs and patients than in that of normal humans and patients( P <0.05~0.01).Conclusion:Th1 and Th2-mediated immunity and IL-4 induced humoral immunity might be the characteristic of immunologic rejection to Xeno-ADM.MLC with sensitized lymphocyte may reflect the in vivo immunoreactivity more accurately.
