摘要
应用国产的聚乙二醇(PEG)、二甲基亚砜(DMSO)、植物血凝素(PHA)和甘油作为融合剂诱导人类食管癌上皮细胞系(Eca-109)和中国田鼠卵巢细胞系(CHO)在悬液中融合。研究了PEG的分子量和浓度,DMSO、PHA和甘油的浓度,两亲本细胞的比例,细胞的总数量,融合后接种培养的密度对细胞融合率的影响。结果表明:两亲本细胞以1:1混合,用40%的分子量为1000的PEG诱导融合,之后以1×10~5个细胞/ml的密度接种培养,获得了最高的融合率。DMSO、PHA和甘油均可加强PEG诱导融合的活力。PHA毒性较强,尤其是浓度高、作用时间长时更甚。此外,甘油本身可诱导融合获得比PEG更高的融合率。甘油作为融合剂尚未见报道。细胞的总数量(在1.6×10~6~1×10~7个之间)和PEG溶液之PH值(在5~9之间)对细胞融合率无影响。
The home-made polyethylene glycol(PEG), dimethyl sulfoxid (DMSO), glycerin
and phytohemagglutinin (PHA) were used, in the present experiment to induce the
fusion of the epithelial cell line of human esophageal cancer (Eca-109) with the
chinese hamster ovary cell line(CHO), in suspension. The effects of the molecular
weight and the concentration of PEG, the concentration of DMSO, PHA and gly-
cerin, the ratio of the two parent cell lines, the total number of the cells, and the
planting density for culture after fusion on the cell fusion (fusion index) were in-
vestigated. The results indicated that the highest fusion index could be obtained
when the 2 parent cell lines were mixed at the ratio of 1 to 1, the fusion was
induced by 40% PEG with the molecular weight of 1000, and then was cultured
with a planting density of 1×10~5 cells/ml after fusion. DMSO, PHA and glycerin
can all enhance the ability of PEG to induce fusion. PHA has high cytotoxicity.
And what is more, glycerin itself can induce cell fusion with the higher fusion in-
dex than that mediated by PEG. There has been no report about glycerin as a fu-
sogen. The total number of cells(from 1.6×10~6 to 1×10~7) and PH of PEG solution
(from 5 to 9) have no effect on the fusion index. These results will be condu-
cive to the study in which the fusion technique of cells in suspension is utilized.
