摘要
采用聚乙二醇法,电场法和改进的聚乙二醇加电脉冲法诱导侧耳属细胞种间融合.用5个强度为10kV/cm,宽度为20μs的高压方波电脉冲处理经7%聚乙二醇(M.W.6000)作用凝集的两亲本原生质体,获得的融合物经双抗性遗传标记和结菇实验筛选,最终得到6株稳定的融合子.6株融合子与双亲有明显差异.实验结果表明,改进后的融合法对侧耳属细胞种间融合是一种有效的方法,具有较高的融合率和再生率,分别达到3.4×10^(-3)和5.6×10^(-3).
Electrically fused interspecific protoplast of the Pleurotus (Fr.) Kumm was achieved by Model CD-300 pulse cell fusion system. Fusion was induced by pulses (10kV/cm, 20μs duration) after the protoplasts lad been collected by PEG, and the fusion frequency of double-anti ability of fusants was 3.4×10-3, as well as the regeneration frequency was 5.6×10-3. It is proved that the i'usant made evidently different of their parental strains with antagonism and acid-phosphatase isozyme.The results show that the electric field-induced protoplast fusion under PEG acting on is an effective method to bring about much high fusion frequency.
