基于数学模型分析不同产地佛手的化学成分差异性 被引量:8

Analysis of chemical diversity of Fructus Citri Sarcodactylis from different habitats based on mathematical model

摘要 目的:通过对6个不同产地的佛手药材进行定性、定量分析,结合数学模型,找出不同产地佛手药材的差异性。方法:采用Kromasil C_(18)色谱柱(4.6 mm×250 mm,5μm),以乙腈为流动相A,0.5%醋酸水溶液为流动相B,进行梯度洗脱,检测波长284 nm,柱温25℃,流速1.0 mL·min^(-1),进样量10μL。对佛手药材中的主要成分—橙皮苷进行定量测定,同时建立了佛手药材的特征图谱。采用ChemPattern型软件构建数学模型,对测定数据进行多元统计分析。结果:以外标法测定18批不同产地的佛手样品中橙皮苷的含量范围为0.019~0.161。特征图谱分析确定了6个共有色谱峰。浙江、福建、四川、广东、广西产的佛手与共有模式比较,相似度在0.85以上,云南产佛手与共有模式比较相似度在0.80以下。数学模型分析结果显示,广东、广西、四川产佛手相距较近。浙江、福建、云南产佛手在建立的模型中与广东、广西、四川的样品相距较远。结论:不同产地的佛手药材,其橙皮苷含量差异明显,通过相似度分析和数学模型统计分析特征图谱数据,可以很好地区分不同产地的佛手药材。 Objective:To distinguish Citri Sarcodactylis Fructus from different habitats by qualitative and quantitative analysis coupled with multivariate statistical analysis.Methods:Determination of hesperidin in Fructus Citri Sarcodactylis and characteristic chromatogram of Fructus Citri Sarcodactylis was performed on a Kromasil C18(4.6 mm×250 mm,5 μm)column with gradient elution of acetonitrile and 0.5% acetic acid at a flow rate of 1.0 mL·min^-1.The column temperature was 25 ℃.Multivariate statistical analysis was performed by ChemPattern software.Results:The contents of hesperidin in 18 batches of Citri Sarcodactylis Fructus were determined by external reference method.The content range of hesperidin was 0.019-0.161. Six common chromatographic peaks were set for similarity analysis and multivariate statistical analysis.The similarity degrees of the samples from Zhejiang Province,Fujian Province,Sichuan Prvince,Guangdong Province and Guangxi Province were above 0.85 compared with common model of characteristic chromatogram;the similarity degree of samples from Yunnan Province was below 0.80.The results of multivariate statistical analysis showed that the samples from Guangdong Province,Guangxi Province and Sichuan Prvince were close to each other,and the samples from Zhejiang Province,Fujian Province and Yunnan Province were far from the above samples.Conclusion:The results from this study indicate that the contents of hesperidin in Citri Sarcodactylis Fructus from different producing areas are different obviously.Qualitative and quantitative analysis coupled with multivariate statistical analysis can be used to distinguish Citri Sarcodactylis Fructus from different habitats.
