摘要
探讨同源异形框基因Six1在结肠癌组织中的表达及其对结肠癌细胞迁移能力的影响.方法 采用免疫组织化学法检测90例结肠癌患者肿瘤细胞及癌旁正常组织中Six1的蛋白阳性表达率.采用针对Six1的siRNA 、niRNA及质粒载体转染结肠癌细胞系 HT-29细胞,RT-PCR法及Western blot法鉴定其转染效果,采用细胞侵袭实验观察各组细胞迁移能力变化.结果 Six1蛋白在结肠癌组织中的阳性表达率显著高于其在癌旁正常组织中的阳性表达率( P<0 .05) .在体外转染实验中,Six1-siRNA组Six1 mRNA 、Six1蛋白表达水平显著低于Six1-niR-NA组及对照组(P<0 .05). Sixl-siRNA组HT-29细胞穿膜细胞数显著少于Six1-niRNA组及对照组(P<0 .05).结论 Six1表达与结肠癌发展密切相关,抑制Six1的表达可以抑制结肠癌细胞的迁移能力.
Objective To investigate the expression of Six1 in colon cancer and its effect on cell migration ability of colon cancer cells.Methods T he positive expression rate of Six1 in 90 cases of colon cancer and adjacent normal tissues were detected by immunohistochemistry method.SiRNA ,niRNA and plasmid vectors were used for transfection of colon cancer cell line HT-29 cells ,RT-PCR and Western blot methods were used to identify the transfection effect.Cell invasion assay was used to observe the changes of cell mi-gration ability.Results The positive expression rate of Six1 protein in colon cancer tissue was higher than that in normal adjacent mucosa (P<0.05).The number of transmembrane cells of HT-29 cells in Sixl-siRNA group was significantly lower than that in Six1-niRNA group and control group (P<0.05).Conclusion The expression of Six1 is closely related to the development of colon cancer ,the inhibition of the expression of Six1 can inhibit the migration ability of colon cancer cells.
