辽宁省大连市种公猪伪狂犬病血清抗体与精液病原的对应检测

Corresponding Detection and Analysis on Serum Antibodyies and Semen Pathogen of Pseudorabies in Breeding Boars in Dalian City of Liaoning Province

摘要 为掌握大连市种公猪伪狂犬病(PR)免疫和野毒感染情况,以及种公猪带毒对猪场PR净化的影响,采用ELISA试验、荧光PCR试验,对24个种猪场的108头种公猪,对应检测伪狂犬病毒(PRV)gE抗体、gB抗体和精液中的PRV核酸,同时检测同场经产母猪的PRV gE抗体、gB抗体。根据种公猪检测结果,划分不同PR净化等级种公猪场,并分析不同等级种公猪场经产母猪的PRV gE抗体。结果显示:种公猪PRV gB抗体阳性率为71.3%,gE抗体阳性率为37.0%;精液PRV核酸阳性率为2.8%。精液PRV核酸检测全部为阴性的一级种公猪场,占比为25%;一级种公猪场的经产母猪PRV gE抗体全部为阴性,二、三、四级种公猪场的种公猪与经产母猪同步呈现从低到高的PRV野毒感染水平。结果表明:种公猪在PR净化中起关键性作用;对于PR净化,对种公猪不但要进行PRV gE抗体检测,还要进行精液病原检测。 In order to recognize tile immunization of PRV vaccine, infection status of wild virus and the influence of breeding boars carrying PRV on disease eradication, 108 breeding boars from 24 pig farms were selected, then by ELISA and fluorescent PCR methods, the antibodies against PRV gB and gE and PRV nucleic acids in semen were detected. Meanwhile, the gE and gB antibodies in multiparous sows in the same farm were also detected. According to detection results of breeding boars, different grade farms for PR eradication were divided, and gE antibodies of sows in different grades of breeding boar farms were analyzed. Results showed that, for the breeding boars, the positive rate ofPRV gB antibody was 71.3%, the positive rate ofgE antibody was 37.0%, and the positive rate of PRV nucleic acids in semen was 2.8%. The proportion of grade I farms (The positive rate of PRV gE antibody in all the breeding boars was 0, and the positive rate ofgB antibody was 100%. PRV nucleic acid couldn't be detected from the semen of soars) was 25%. In the farms of grade I, the gE antibodies couldn't be detected from multiparous sows. Otherwise, the infection level of wild PRV showed a trend of low to high in breeding boars and multiparous sows in farms of grade II, III and IV. As a conclusion, the breeding soars played a key role in PR eradication. As to PR eradication, both the gE antibody and PRV DNA in the semen should be detected.
