摘要
目的:建立烟叶中茄呢醇的含量测定方法,为其提供质量标准。方法:应用高效液相色谱-蒸发光散射检测仪,Discovery^(TM)C_(18)(25cm×4.6 mm)色谱柱,以甲醇-乙腈-四氢呋喃(39:24:9.5)为流动相,流速为1.0mL·min^(-1),测定烟叶中茄呢醇的含量。结果:分析了山东、四川等地产烟叶及其提取物中茄呢醇的含量,不同产地烟叶中茄呢醇的含量为0.134%~0.373%,自制提取物中的含量分别为74.36%和91.02%。加样回收率为97.6%(RSD=2.0%)。结论;该方法快速、简捷、重现性好,可以用于烟叶中茄呢醇的质量控制。
Objective: To establish a method to determine the contents of solanesol in Nicotiana tobaccum L. , and to evaluate the quality of these herbs. Methods: With evaporative light scattering detection (ELSD) , a Discov-eryTM C18 (25 cm×4. 6 mm) as analytical column, methanol - acotonitrile - tetrahydrofuran (39:24:9.5) as the mobile phase, and a flow rate of 1.0 mL ·min-1 , the contents of solanesol were measured. Results: Several kinds of samples of Nicotiana tobaccum and their extracts were analyzed; The contents in herbs from different areas were 0. 134% -0. 373% , and the contents in their extracts were 74. 36% and 91. 02%. The mean recovery of solanesol was 97. 6% (RSD = 2. 0% ). Conclusion: This method is fast, simple and reproducible, and it can be used for the quality control of Nicotiana tobaccum.
