摘要
【目的】研究地理因素对水蛭素基因结构变异的影响。【方法】抽提广东菲牛蛭基因组DNA ,根据已发表的水蛭素基因设计一对引物 ,从基因组DNA扩增出一特异性的大约 70 0bp片段 ,再回收、克隆和测序。【结果】广东菲牛蛭水蛭素基因全长为 6 42bp ,基因有 4个外显子 ,被三个内含子隔开。与马尼拉菲牛蛭水蛭素基因Hm1、Hm2 的同源性分别为 90 %与和 88 6 %。【结论】地理因素会影响菲牛蛭水蛭素基因结构的变异。
Objective To study the effect of geological factors on hirudin gene variation in construction Methods The genomic DNA were isolated from Hirudinaria manillensis living in Guangdong. A pair of primers was designed from published hirudin genes. A specific DNA segment about 700 bp full-length was obtained from genomic DNA amplification by PCR. The segment was retrieved?purified?cloned and sequenced. Results The hirudin gene from Guangdong Hirudinaria manillensis consisted of 640 bp. The gene contained four exons interrupted by three introns. The gene sequence had a homology with Hm 1 of 90%, Hm 2 of 88.6% from Manila Hirudinaria manillensis.Conclusion The geological factors have an effect on hirudin gene variation of Hirudinaria manillensis.
