摘要 报道了水蛭素全基因的合成及克隆。根据已知水蛭素氨基酸序列及酵母菌体内蛋白质表达特性 ,设计了水蛭素的全基因序列 ,采用化学合成及酶学相结合的方法 ,合成了该基因片段 ,并逐段克隆至载体 pBS上 ,得到全长水蛭素基因。序列测定表明 ,克隆后DNA顺序同原设计序列。 A 240-bp DNA fragment coding for the hirudin was chemically and enzymically sythesized.The sequence was derived from the amino acid sequence of the hirudin peptide,which is consistent with the request of yeast gene expression.The sythesis involved preparation of six long oligodeoxyribonucleotides.After annealing,filling by Klenow fragment,three formed fragments were cloned into pBS sequentially,and an intact hirudin gene was obtained.The DNA sequence of the cloned fragment identified by sequencing was the same as the designed sequence of hirudin gene
