抗人血型A抗原单克隆抗体的研制

摘要 用细胞融合法获得两株稳定分泌抗人血型A抗原单克隆抗体的杂交瘤细胞株(HA_3和HA_4)。抗体特异性很好,与“O”和“B”型红细胞无交叉凝集,千余份血型测定结果与标准血清一致;抗体为IgM类,耐热、耐冻融,可望代替标准血清供市售生产。 <ABSTRACT> Using cell fusion technique two hybridoma cell lines secreting specific monoclonal antibodies (McAbs)against human blood group A antigen(HA_3 andHA_4) were established. The specificity of these two McAbs is satisfactory.They didn′t agglutinate with type“O”or“B”of human blood red cells. More than a thousand human blood samples had been examined with these twoMcAbs. The results were well consistent with that of standard serum. Both HA_3 and HA_4 McAbs were of the class IgM, resistant to heat and freezingthawing. If be produced commercially, it is possible to replace the standardblood group typing serum.
