大肠癌中NF-κBp65、VEGF表达的相关性研究 被引量:10

摘要 [目的]检测细胞核因子κB(NF κB)、VEGF在大肠癌组织中的表达并探讨其与大肠癌肿瘤血管生成关系。[方法]应用免疫组织化学方法检测NF κBp65及VEGF在52例大肠腺癌组织、21例大肠腺瘤及10例正常大肠组织中的表达情况 ,同时观察肿瘤内微血管密度 (MVD)。[结果]52例大肠腺癌中34例NF κBp65阳性 ,阳性产物主要定位于肿瘤细胞胞浆 ,21例大肠腺瘤中10例NF κBp65阳性 ,10例正常大肠组织中NF κBp65无1例阳性表达 ,NF κBp65表达与患者的性别、年龄、大肠癌组织学分型、淋巴结转移、Dukes′分期无关 ;VEGF阳性表达率为61.5% ;MVD在NF κBp65阳性大肠癌中明显高于阴性者 ;NF κBp65表达与VEGF、MVD表达呈显著正相关。[结论]NF κBp65是一个大肠癌相关的癌蛋白 ,NF κBp65对VEGF可能有正向调节作用 。 To investigate the expression of NF κB p65 protein,VEGF in the colorectal carcinoma and relationship to the tumor angiogenesis.The expression of NF κB p65 protein,VEGF protein expression and MVD in tissues from in 52 cases with colorectal carcinoma, 21 cases with large intestine adenoma and 10 from normal colon tissue was studied immunohistochemically.Thirty_four of 52 colorectal carcinoma specimens were positive for NF κB p65 protein with a 65.4% positive rate ,10 cases were found to have NF κB p65 protein positive stain in 21 cases large intestine adenoma, In contrast, none of the normal tissues exhibited NF κB p65 protein (P<0.01). The NF κB p65 protein expression was not correlated to the sex, the location of carcinoma, histological grade, lymph node metastasis or Dukes′ staging (P>0.05).The positive rate of VEGF expression was 61.5%;MVD was significantly higher in NF κB p65 positive CRC than that in NF κB p65 negative CRC (P<0.01).The expression of NF κB p65 was closely related with the expression of VEGF and MVD.[Conclusions]NF κB p65 is an oncogenic protein related to the colorectal carcinoma. NF κB p65 may have positive regulation on VEGF, and influence the tumor angiogenesis.
