摘要
发根农杆菌是一种寄主非常广泛的土壤细菌,能够侵染几乎所有的双子叶植物和少数单子叶植物。在自然状况下,发根农杆菌通过伤口主要侵入植物的茎基部,导致不定根和侧根的大量发生,以致出现毛根病的症状。在此过程中,发根农杆菌对植物器官、组织或细胞的侵染伴随着其RiT-DNA向植物基因组内的转移。
Strain LBA9402Bin19 of Agrobacterium rhizogens, harboring binaryvector of Bin19, incited roots in vitro on hypocotyl segments of cab-bage, oilseed rape and cauliflower. Among these roots, the transformedroots containing Kn^r gene were resistant to kanamycin and grew on themedium containing 50mg/L kanamycin,whereas untransformed roots werenot resistant to kanamycin and abandoned. Among three genotypes of Brassica crops tested, cauliflower had thehighest root induction percentage while cabbage produced the highestroot transformation frequency. Within the population of 50 transgeniclines, The introduction of Kn^r gene from binary vector Bin19 was notalways synchronized with that of ags and mas genes from Ri T-DNA.
