摘要
肿瘤坏死因子(TNF)对许多肿瘤细胞具有细胞毒和抑制生长的作用。为进一步研究TNFα的结构和功能,并为临床研究提供rHTNFα纯品。本实验应用杂交瘤技术制备了一组抗rHTNFα单克隆抗体(T_5、S_3、H_7、B_3、B_5、E_6、Z_(12)、Z_8和Z_(20))。ELISA结果证实它们与rIL-1、rIL-2、rIFNγ、rIFNα_1及E.coli菌裂解液(MM_(294))无均交叉反应,仅与rHTNFα有反应。Western blot结果进一步证实这组抗体对rHTNFα的特异性。这组抗体具有不同程度中和rHTNFα细胞毒能力。用抗rHTNFα抗体制备的亲和层析柱纯化rHTNFα,可以得到在SDS-PEAG上分子量为17000道尔顿的rHTNFα纯品。
Tumor necrosis factor (TNFα)plays an important role in cytotoxicityand inhibition of tumor cells. Further studies on the structure, functionand clinical application of TNFα will be useful. Nine clons of hybridomasecreting monoclonal antibodies against rHTNFα were obtained by usingcell fusion technology. None of the monoclonal antibodies cross-reactedwith rIL-1, rIL-2, rIFNγ, IFNα_1, and E.coli lysates. Western blotdomonstrated that they specifically recognized rHTNFα antigen of M.W. 17000 daltons. Some of the antibodies recognized native rHTNFα,too.These antibodies neutralized the cytotoxicity of rHTNFα to differentextents. They will be utlized as imimunoaffinity column to purifyrHTNFα from recombinant E. coli lysates.
