青霉素G钾在仔猪体内代谢动力学研究

On Pharmacokinetics of Penicillinum G Kalium in Piglets

摘要 分别选用供试初生未哺乳及3日龄哺乳的杜长杂交仔猪各8头及长本杂交仔猪各4头,以10万HU/kg体重的剂量,给两类仔猪一次内服青霉素G钾。在给药后的24小时内定时采血,以滕黄八叠球菌为供试菌种,按中国药典抗生素微生物检定法进行抑菌试验,测得的血药浓度与时间数据,按MCPKP—自动化药动学分析程序进行分析,数据符合一级吸收二室模型,分别得出两类仔猪各自的指数方程和内服本剂的主要药动学参数。表明两类仔猪一次内服本剂后体内分布并不规律;仔猪品种间及个体是否哺乳间差异均较大。其达峰时间(Tmax)为0.73±0.06小时,生物半衰期(t1/2β)为0.91±0.02小时,有效杀菌浓度维持时间(Tcp)为4.31±0.11小时. Two kinds of hybrid piglets, sixteen Duroc with Landrace (DXL)and eight Landrace with Local (LXL), were selected for experiment. Each kind of piglets was equally divided into two groups. Newborn piglets without feeding and three-days piglets with feeding.Penicillinum G Kalium was given to piglets with a dose of 10×10~5 I. U./kg body weight. The blood samples were collected at fixed time within twenty-four hours after the drug was given. According to pharmacopoeia antibiotic microbe test, with sarcina lutea as a vaccine, the data of blood concentration and time were obtained and analysed with the MCPKP-Auto Pharmacokinetical Software. The result corresponds with the two compartment open model of first order absorbtion. Also an index equation and main parameters of the two kinds of piglets have been got. The experiment showed that the distribution of drug in test piglets had no regularity, and there were significant differences between breeds and between fed and non-fed piglets. T_(max): 0.73±0.06 h, T_(1/2)β: 0.91±0.02h, T_(cp): 4.31±0.11 h.
