摘要 目的 :建立荧光分光光度法测定人脑脊液中左氧氟沙星浓度的新方法。方法 :以三氯乙酸为蛋白沉淀剂 ,加醋酸 醋酸钠缓冲液 (pH4.0 )后测荧光强度。结果 :激发波长为 2 94nm ,发射波长为 496nm ,浓度在 0 .1~ 5 .0μg/ml范围内线性关系良好。标准曲线回归方程为Y =47.482 6X +2 .85 2 4,相关系数r=0 .9998,平均相对回收率为 98.2 % ,变异系数为 2 .16 % ,结论 :本方法操作简便 。 Abstract:To establish a spectrofluorometry for determination of levofloxacin(LVFX) concentration in human cerebrospinal fluid(CSF). Methods: Human CSF protein was precipited with trichloroacetic acid and fluoroscent intensity of LVFX was measured in acetic acid sodium acetate buffer(pH4.0). Results: Its λ ex and λ em was 294 nm and 496 nm, respectively. Linear relationship was found in the range from 0.1~5.0 μg/ml. The regression equation was Y= 47.4826X +2.825 4. The correlation coefficient(r) was 0.999 8. The average recovery was 98.2%. The coefficient of variation was 2.16%. Conclusion:The method is ripid, simple and suitable for determination of LVFX in human CSF.
