摘要
目的 :研究土槿甲酸 (PAA)对人红白血病 (K5 6 2 )细胞生长的抑制效应 ,探讨其抗癌作用机制。方法 :用不同浓度的 PAA加入体外培养的 K5 6 2细胞中 ,观察加药后细胞生长和有丝分裂指数 (MI)的变化 ;用 MTT法检测其细胞毒作用 ;Hoechst33342 /PI荧光双染色方法检测细胞凋亡。结果 :PAA明显抑制 K5 6 2细胞生长 ;IC50 值为5 0μmol/L ;MI于加药后 2 4小时较对照组明显降低 ;凋亡细胞比例在用药后 2 4小时达 80 %。结论
Objective: To study the effects and its mechanism of pseudolaric acid A on Proliferation of K562 cells. Methods: Using experiment in vitro, pseudolaric acid A of different concentrations were into the medium in which the cells were cultured. The changes of cell proliferation were assessed by MI and cell growth number. Cytoxicity rate was detected by MTT. The number of apoptotic cells was quantitatively assessed by Pseudolaric acid A, the IC50 values of Pseudolaric acid A for K562 cells was 50μmol/L; after 24h of exposure to Pseudolaric acid A, MI was remarkably reduced, the rate of Apoptotic cells was increased. Conclusion: Malignant phenotype of K562 cells was changed and its proliferation ability was remarkably inhibited by Pseudolaric acid A.
