摘要
目的 进一步研究β2 糖蛋白Ⅰ (β2 GPI)、心磷脂 (CL)和抗心磷脂抗体 (aCL)三者之间的关系 ,以及 β2 GPI在aCL致血栓形成中所起的作用 ,从人血清中提纯 β2 GPI。方法 采用高氯酸沉淀血清中的杂蛋白 ,过DEAE纤维素层析柱和DEAESephadexA 5 0层析柱。 结果 经SDS PAGE测定纯化样品的分子量为 5 0KD ;经皮免疫小鼠后 ,小鼠血清中可检测到高滴度的aCL。结论 上述组合方法方便可行 ,为β2 GPI的进一步研究奠定了良好的基础。
Objective To purify β 2-glycoprotein Ⅰ(β 2-GPI) from human serum in order to find out mechanism of β 2-GPI in thrombosis and the relationship of β 2-GPI, cardiolipin(CL) and anticardiolipin antibody(aCL). Methods β 2-GPI was precipitated with perchloric acid, this treatment was followed by ion-exchange chromatography on DEAE fibrin and DEAE Sephadex A-50.Results The molecular weight(50?KD) of sample was characterized by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. When the mice were immunized with the sample, there was high titre of aCL in serum.Conclusion The method above-mentioned is a feasible way. It can creat a favorable foundation for more about β 2-GPI.
