摘要
目的 :观察类风湿关节炎大鼠模型肺脏不同时间的病理变化、血小板源性生长因子 (PDGF)及其受体的表达 ,探讨风湿肺发病机制。方法 :Wistar大鼠用福氏完全佐剂致类风湿关节炎 (RA)后 ,选择 36只分别在不同时期和应用氢化考的松及低分子量肝素进行治疗后 ,取其肺脏观察HE染色病理变化及免疫组化染色检测PDGF配体及受体表达情况。结果 :致炎模型 15d时肺泡炎最严重 ,30d时肺纤维化最严重。PDGF配体及受体在正常肺组织中表达较少 ,RA 15d时表达高峰 ,RA 30d时有所回落。氢考和低分子量肝素治疗后明显下降。结论 :风湿肺的变化经历了从肺泡炎至肺纤维化的过程。PDGF配体及受体表达增高与风湿肺病理改变加重一致。低分子量肝素治疗风湿肺与氢化考的松具有同样作用 。
Objective: Our aim was to observe the pulmonary pathological changes of rheumatic rat models and the expression of platelet derived growth factors (PDGF) and their receptors on the lungs. We also investigated the mechanism of rheumatic lung. Methods: Thirty six rheumatoid arthritis(RA) Wistar rat models by Freund's complete adjuvant were selected. Then the pathological changes of lungs with HE staining in different periods or treated with hydrocortisone and low molecular weight heparin, and PDGF and receptors expression with immunohistochemistry were demonstrated. Results:The pathological changes of RA rats in different periods were showed. Alveolitis were most severe in lungs of 15 days, while pulmonary fibrosis were most severe in lungs of 30 days. The PDGF and their receptors expressed little in healthy lungs. They expressed more on the lungs of RA 15 days than that of 30 days. The expression of PDGF decreased significantly with administrating low molecular weight heparin as same as hydrocortisone. Conclusion:The changes of rheumatic lungs were from alveolitis to fibrosis. The higher were expression of PDGF and their receptors, the more severe were pathological changes. Low molecular weight heparin had same effects on treatment of rheumatic lungs as hydrocortisone. Rheumatic lungs be controlled by PDGF inhibitor.
