血小板源性生长因子及其受体在类风湿性关节炎大鼠肺组织分布及对血气影响

摘要 目的 :观察血小板源性生长因子 (platelet derivedgrowthfactor,PDGF)及其受体在类风湿性关节炎的大鼠模型肺组织分布及对血气影响。方法 :Wistar大鼠用福氏完全佐剂致类风湿关节炎后 ,选择 2 4只分别在不同时期取其肺脏进行免疫组化染色检测 ,观察PDGF配体及受体分布情况。同时采大鼠腹主动脉血用瑞士ALVOMINI5血气分析仪检测。结果 :免疫组化染色最强的细胞为增厚的肺间质细胞和肺血管周围成纤维细胞。PDGF配体及受体不仅在血管平滑肌细胞中有表达 ,而且在内皮细胞 巨噬细胞中均有表达。PDGF阳性细胞数与血氧分压 pH值呈负相关 ,与二氧化碳分压呈正相关。结论 :PDGF通过自分泌和旁分泌方式刺激风湿肺内各种细胞的增殖变化 。 Objective: Our aim was to observe the distribution of platelet derived growth factors (PDGF) and their receptors on the lungs of rheumatic rat models and the effects on blood gas analyses. Methods: Twenty four rheumatoid arthritis Wistar rat models by Freund's complete adjuvant were selected. Then the PDGF and receptors distribution with immunohistochemistry were demonstrated. Arterial blood gas values were measured via Swiss AVL OMINI5 blood gas analysis instrument. Results: The most intense immunohistochemistry stain cells were pulmonary interstitial cells and fibroblast around blood vessels. The expressions of PDGF and their receptors were not only on smooth muscle cells of blood vessels but also on endothelial and macrophage cells. The positive cells of PDGF were negatively correlated with PaO 2, pH values, and were positively correlated with PaCO 2 values. Conclusion: The PDGF stimulated several kinds of cells in the lungs in growing and changing by self secreting and side secreting, and had effects on blood gas.
