激活的高分子激肽原抗细胞伸展与体外连接蛋白结构变化关系的探讨 被引量:2

The Relationship Between Anti-Cell Spreading of Kinin-Free High Molecular Weight Kininogen and the Conformational State of Vitronectin

摘要 目的 :通过观察结构上改变的体外连接蛋白 (aVN)底物上激活的高分子激肽原 (HKa)对细胞伸展的影响 ,比较VN与aVN上HKa对细胞抑制活性。方法 :采用细胞杂交及细胞粘附方法观察在HKa存在与否下VN对细胞伸展的影响。结果 :HKa存在下MG 6 3细胞不能在固定的VN上伸展 ,但能在aVN上伸展 ;天然VN固定于塑料表面后用尿素处理 ,HKa存在或缺乏状态下细胞伸展活性相同。细胞杂交结果显示 ,在包被VN的硝酸纤维素膜上MG 6 3细胞伸展 ,存在HKa状态下细胞不能伸展于天然VN ,而伸展于aVN底物。结论 :在天然VN与aVN底物上HKa对细胞的抑制活性不同 。 Objective: This paper was designed to analyze the effects of kinin free high molecular weight kininogen (HKa) on cell spreading to altered vitronectin(aVN). Methods: The cell spreading was observed by cell adhesion assay and cell blotting system. Results: HKa did not show the anti cell spreading activity when altered vitronectin was used as a substratum. The MG 63 cell attached very well on the native vitronectin. Cell did not attach to the native vitronectin in the presence of HKa, but attached to the 65 kDa molecule in aVN even in the presence of HKa. Conclusion:The anti cell spreading effects of HKa are also modulated by the labile conformation of substratum.
